Orforglipron is a once-daily oral GLP-1 weight loss pill (just like Wegovy Pill) developed by Eli Lilly, sold in the UK under the brand name Foundayo. It was approved by the FDA on 1 April 2026 and is now approved in the UK as of Monday 10th August by the MHRA.

What Is Orforglipron?
Orforglipron is another GLP-1 receptor agonist, like semaglutide (Wegovy). That means it works by mimicking a gut hormone called GLP-1 that your body naturally releases after eating. It tells your brain when you’re full, slows down how quickly food leaves your stomach, and helps regulate blood sugar.
Orforglipron is basically a synthetic version of this hormone, with amplified effects. It replicates this effect continuously, which is why appetite reduces, food noise quietens, and sustained weight loss becomes possible.
However, orforglipron differs from other GLP-1 medicines in its chemistry (and the difference is game changing).
Semaglutide (the active ingredient in Wegovy) and tirzepatide (in Mounjaro) are peptides, i.e., large protein based molecules that must be injected or prepared in special oral formulations with strict fasting requirements.
Orforglipron is a small molecule. It is chemically simpler, survives stomach acid without any special technology, and absorbs reliably regardless of food intake or timing.
Who Makes It, and Where Does the Name Foundayo Come From?
Eli Lilly, the same company that makes Mounjaro (tirzepatide) developed Orforglipron. Foundayo is the name chosen by Lilly for the US commercial launch.
People use both names used interchangeably online. For UK searches and clinical discussions, "orforglipron" is the more commonly used term at this stage, since Foundayo is specifically the US trade name and we don’t have an official UK name yet.
What Is Orforglipron Used For?
In the UK, orforglipron has been authorised for weight loss and weight maintenance in adults with a BMI of 30 or higher, or a BMI between 27 and 30 alongside at least one weight-related comorbidity. It has also been authorised to support glycaemic control in patients with type 2 diabetes that is not adequately controlled.
How Does Orforglipron Work?
Orforglipron attaches itself to GLP-1 receptors in the brain and gut, and produces the same results as injectable GLP-1 medicines. You’ll see appetite reduction, slower gastric emptying, more satiety, and better blood sugar control. With time, your body’s weight set point gradually shifts downward.
A Small Molecule, Not a Peptide: What That Means in Practice
This is where Orforglipron brings the game changing distinction.
Semaglutide is a peptide-based drug. Peptides are proteins that are broken down by stomach acid and don’t get absorbed through the gut lining easily. Hence, for it to work as an oral medicine, the Wegovy Pill contains something called SNAC (the full form is a bit difficult to pronounce, so let’s just stick to SNAC). It temporarily modifies the stomach environment to allow the semaglutide through. But for SNAC to work, the stomach needs to be completely empty. This is why the Wegovy Pill comes with strict rules of fasting, exactly 120ml still water and a 30-minute wait before eating or drinking anything else.
Orforglipron is not a peptide. It is a small molecule, built to be taken orally without any of those constraints. It survives stomach acid on its own, absorbs through the gut lining reliably, and hence doesn't require a special carrier like SNAC or strict timing protocol.
No Food or Water Restrictions: How It Differs From Other Oral GLP-1s
Orforglipron comes with no restrictions. Any time of the day. Any amount of water. With or without food. No morning rule. No 30-minute fasting.

Orforglipron Weight Loss Results: What the Trials Showed
Two large Phase 3 trials provide the core evidence for orforglipron's weight loss effects.
ATTAIN-1: Adults With Obesity or Overweight, Without Diabetes
ATTAIN-1 enrolled adults with obesity or overweight and at least one weight-related health condition, but without type 2 diabetes. The trial ran for 72 weeks and tested three doses: 6mg, 12mg, and 36mg daily.
- At the highest dose (36mg): average weight loss of 12.4% of body weight at 72 weeks
- 59.6% of participants on the 36mg dose lost at least 10% of their body weight
- 39.6% lost at least 15%
- Participants saw significant improvements in waist circumference, blood pressure, and cholesterol
- Among participants with prediabetes at baseline: up to 91% achieved near-normal blood sugar levels, compared to 42% on placebo
ATTAIN-1 results were presented at the European Association for the Study of Diabetes (EASD) Annual Meeting in September 2025 and simultaneously published in the New England Journal of Medicine.
ATTAIN-2: Adults With Type 2 Diabetes
ATTAIN-2 was done specifically to test orforglipron in adults with obesity or overweight and type 2 diabetes. This is a group typically harder to achieve weight loss in because diabetes itself alters metabolism and many diabetes medications promote weight gain.
- At the highest dose (36mg): average weight loss of 10.5% at the primary endpoint
- HbA1c (a measure of blood sugar control) reduced by an average of 1.8 percentage points
- 75.5% of participants achieved HbA1c below 7%, a common treatment target
These results were reported in August 2025, ahead of the FDA approval submission.
There was an interesting third trial done as well: ATTAIN-MAINTAIN, published in Nature Medicine in May 2026. It enrolled women and men who had previously taken injectable GLP-1 medicines (tirzepatide or semaglutide) and then switched to orforglipron.
The study found that orforglipron successfully maintained approximately 74.7% of the weight reduction that had been achieved on the injectable treatment, suggesting it may be a practical oral continuation option for people who switch off injectables.
How These Results Compare With Current Options
Orforglipron's 12.4% weight loss at 72 weeks (ATTAIN-1, 36mg) is similar to 13.6% for oral semaglutide at 64 weeks (OASIS 4) and 14.9% for injectable semaglutide at 68 weeks (STEP 1). Mounjaro injection (tirzepatide), however, produces much higher average results at 22.5% at 72 weeks (SURMOUNT-1).
So the trade-off with orforglipron is not efficacy versus injections, it is convenience versus efficacy within the oral format, and ease of use versus the Wegovy pill specifically.

Orforglipron vs Other GLP-1 Options
The most direct comparison for orforglipron is only possible with the Wegovy pill since both are once-daily oral medicines. Both are genuinely needle-free, both work through the same GLP-1 mechanism, and both require prescription and clinical oversight.
The key difference, we’ve already established earlier, is the fasting and timing protocols. While there are some rules to be followed for the Wegovy Pill, orforglipron comes with no restrictions.
In terms of weight loss results, the Wegovy pill edges slightly ahead (13.6% at 64 weeks vs 12.4% at 72 weeks for orforglipron), but head-to-head trial data directly comparing the two in the same study is limited.

Orforglipron Side Effects and Safety
Orforglipron has a similar side effect profile as the other GLP-1 medicines with gastrointestinal effects being the most common, particularly during the dose escalation period.
- Nausea: most pronounced at the start of each new dose level and typically improving after a few weeks
- Diarrhoea, constipation, and vomiting: Similar to what people on other GLP-1 medicines experience
- Decreased appetite: an intended effect, but worth noting that it can lead to reduced caloric intake requiring attention to protein and nutrient intake
⚠️ Boxed warning: thyroid C-cell tumours. Like all GLP-1 receptor agonists, orforglipron carries a boxed warning about the risk of thyroid C-cell tumours observed in rodent studies. This risk has not been confirmed in humans, but orforglipron is contraindicated in people with a personal or family history of medullary thyroid carcinoma or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2). This warning applies to all GLP-1 medicines including Wegovy and Mounjaro.
